The scene in question is the 15th episode of TBBT's 8th season, in which Howard told his friends about his mother's death. Sheldon, who doesn't always know what to say at such moments, approached Howard, mentioned his dad's death and said a touching thing to him: “I didn’t have any friends to help me through it. You do”. So he showed him support in a difficult moment. After these words, Penny said a joke, followed by laughter in the background, and this is what viewers have a big problem with.
Especially since this scene was a farewell not only to Howard's mom, but also to her actress, Carol Ann Susi, who passed away in 2014. Fans of The Big Bang Theory are of the opinion that although it is a comedy series, jokes are not always necessary, and this scene should have been kept in a more serious tone.
